    <p>В связи с широким применением растительных метаболитов в народной медицине, фармацевтической, пищевой и других отраслях промышленности, актуально изучение их токсичности с целью оценки безопасности для человека. Исследования in vivo большого количества образцов на токсичность с использованием животных в качестве тест-организмов являются дорогостоящими, так как требуют значительных материальных и временных ресурсов, а также одобрения этического комитета. В связи с этим востребованы альтернативные методы оценки токсичности объектов исследования, позволяющие сократить количество образцов, которые в дальнейшем будут подвергаться более углубленному изучению (например, на грызунах). Одним из альтернативных подходов оценки токсичности является использование модельных тест-организмов in vitro, например микроорганизмов. Целью данной работы являлась оценка токсичности ряда растительных метаболитов in vitro, с использованием биолюминесцентных бактерий Aliivibrio fischeri, и их влияния на ряд представителей микробиоты ЖКТ.&#13;
Объектами исследования послужили растительные метаболиты: рутин, розмариновая и транс-коричная кислоты, кверцетин, кемпферол, байкалин и вогонин. Их степень чистоты составляла более 94 %. Данные метаболиты выделены в ходе ранее проведенных исследований из каллусных, суспензионных и корневых культур растений Сибирского федерального округа. Токсичность этих соединений определялась с помощью биолюминесцентных бактерий (A. fischeri): оценивалось влияние веществ на интенсивность свечения этих микроорганизмов. Анализ проводился с помощью тонкослойной хроматографии. Дополнительно оценивалось влияние исследованных растительных метаболитов на тест-штаммы: Propionibacterium jensenii (B-6085), Propionibacterium freudenreichii (B-11921), Lactobacillus freudenreichii subsp. freudenreichii (B-6561), Lactiplantibacillus plantarum (B-884), Bifidobacterium longum (AC-1257), Bifidobacterium bifidum (AC-1779). &#13;
Установлено, что растворы рутина, кверцетина, вогонина и байкалина в 20 % этаноле проявляли токсичность по отношению к A. fischeri. Среди рассматриваемых объектов исследования только кемпферол стимулировал прирост биомассы лакто- и бифидобактерий. Кверцетин, рутин и транс-коричная кислота подавляли рост биомассы пропионовокислых бактерий. Остальные вещества подавляли негативное влияние 20 % этанола, не влияя на рост тест-штаммов. &#13;
Результаты проведенного исследования подтвердили целесообразность предварительной оценки токсичности растительных объектов с помощью биолюминесцентных бактерий A. fischeri как промежуточного этапа перед in vivo исследованиями.</p>

   <trans-abstract xml:lang="en">
    <p>Plant metabolites undergo a thorough toxicity test before becoming part of pharmaceuticals or functional food products. In vivo toxicity studies on animals are expensive and time-consuming. Moreover, they require an ethic approval and a lot of expendables. Alternative methods often involve microbial models. As a result, they reduce the number of animal test subjects on further research stages. This study tested the toxicity of several plant metabolites in vitro on Aliivibrio fischeri and gastrointestinal microbiota.&#13;
The research included rutin, rosmarinic acid, trans-cinnamic acid, quercetin, kaempferol, baicalin, and wogonin (≥ 94%). These plant metabolites were isolated from callus, suspension, and root cultures of Siberian plants. Their toxic effects were tested on the bioluminescent properties of Aliivibrio fischeri. The analysis relied on the method of thin-layer chromatography. Another experiment assessed the toxic effects of these plant metabolites on Propionibacterium jensenii (B-6085), Propionibacterium freudenreichii (B-11921), Lactobacillus freudenreichii subsp. freudenreichii (B-6561), Lactobacillus plantarum (B-884), Bifidobacterium longum (AC-1257), and Bifidobacterium bifidum (AC-1779). &#13;
The solutions of rutin, quercetin, wogonin, and baicalin (20% ethanol) were toxic towards A. fischeri. Kaempferol was the only metabolite that stimulated the biomass growth of lacto- and bifidobacteria. Quercetin, rutin, and trans-cinnamic acid inhibited the biomass growth of propionic bacteria. The other metabolites suppressed the negative impact of 20% ethanol without affecting the growth of the test strains. &#13;
A. fischeri tests proved to be a reliable preliminary toxicity assessment of plant materials before in-vivo studies.</p>
